In the positive control group, a change is expected. A positive control was used as a measure of the severity of the difference between the experimental groups. An example of a positive control is a toxin administered to an animal, which results in reproducible physiological changes or damage. Later, you can use new therapies in the experimental group to determine if these changes can be prevented or cured. Positive controls are also used to demonstrate that reactions can be detected and provide some quality control over the experimental method.

Positive control guarantees that the protocol is not disturbed or that the reagents are not degraded. If your positive control gets a negative result, you know that all other negative results in your treatment group should be tested more carefully, and fresh reagents I might use it

Active control transcription activator, catabolism gene activator (CAP) is a positive control of gene expression. In addition to many other inducible operons, CAP also activates transcription of the lac operon. Since glucose is the preferred food source, the lac operon is not activated in E. coli cells cultured in media containing glucose and lactose until glucose is consumed. However, due to the presence of lactose, it can be expected that the lac operon is derepressed and hence active. However, since the experiments show that glucose itself inhibits the activity of the lac operon, it is activated only when lactose is the sole energy source. This glucose inhibition is observed for many other operons encoding enzymes for use with alternative energy sources. Glucose inhibition occurs through a positive mechanism. As glucose is consumed, its level in the cell decreases
