Abstract In order to study gene mutations that should cause Paget 's bone disease, researchers must have feasible candidates to host genetic mutations. They found the best candidate for having genetic mutations in the mouse so they transplanted genetic mutations into embryos of descendants of the mouse. Researchers hypothesized that p62P394L is sufficient to induce PDB, especially since the p62 gene is involved in the coding of a 62 kDa protein that plays a role in the signaling of osteoclast precursors. Results were found by immobilizing the 1st, 5th lumbar spines of 4, 8 and 12 month old homozygotes, heterozygotes and WT littermates for 10 to 4 hours in buffered formalin for 24 to 48 hours .

In 1877, Sir James Paget described the local bone disease first known as Paget's disease. Paget's disease is a metabolic bone disease characterized by excessive bone resorption and formation by activated osteoclasts. Skeletal Paget's disease usually occurs in people over 40 years of age and mainly affects the axis bone. Approximately 5% of patients develop symptoms that require treatment, but the most common symptom is deformation of bone pain, swelling, painful areas. Other signs of this disease include increased susceptibility to fractures, hearing loss, and neurological complications. PDB is the second most common metabolic bone disease after osteoporosis. This is more common in western countries; in contrast, it is extremely rare in most parts of China and sub-Saharan Africa.

Arthralgia and bone lacquer are common symptoms of Paget's disease. As the disease progresses, bone swelling and expansion weakens the expanded bone. The initial symptoms of Paget 's disease are not clear, but over time, as the disease progresses it becomes clearer. Another complicating factor of Peggy's disease is gout. Gout is caused by overproduction of uric acid and salt and causes gouty arthritis, uric acid metabolism disorder. Accumulation of uric acid and salt in blood leads to the accumulation of bone joint / cartilage, then kidney stone. Bone tumor caused by gout, in particular cancellous bone tumor
